Glutathione (γ-L-glutamyl-L-cysteinylglycine) is a low–molecular weight endogenous tripeptide composed of glutamate, cysteine, and glycine. It is widely distributed in mammalian cells and exists primarily in reduced (GSH) and oxidized (GSSG) forms. In laboratory research, glutathione is classified as a central intracellular redox buffer and thiol-containing antioxidant, playing a critical role in maintaining cellular redox homeostasis and protecting against oxidative stress–induced damage.

Mechanistically, glutathione functions through its reactive sulfhydryl (–SH) group on the cysteine residue, which donates electrons to neutralize reactive oxygen species (ROS) and electrophilic intermediates. During this process, two GSH molecules can form glutathione disulfide (GSSG), which may be recycled back to the reduced form via glutathione reductase in an NADPH-dependent reaction. In addition to direct antioxidant activity, glutathione serves as a substrate for glutathione peroxidases and glutathione S-transferases, supporting enzymatic detoxification pathways and regulation of redox-sensitive signaling cascades.

In vitro and preclinical research applications, glutathione is frequently utilized to evaluate oxidative stress markers, mitochondrial function, and redox-regulated gene expression. Investigators may assess intracellular GSH:GSSG ratios as indicators of cellular oxidative status, or examine glutathione’s influence on protein thiol modification, apoptosis pathways, and xenobiotic metabolism. Due to its central biochemical role, it is commonly incorporated into experimental models studying inflammation, toxicology, and metabolic regulation.

Overall, glutathione is regarded as a foundational research compound for studying cellular antioxidant defense systems and redox biology.
